Moriarty , PhD[Associated Requests/Data] Assistant Professor,
University of Toronto Subgroup(s): [Glycans in host-microbe interactions]
Research interests: We study the mechanism of vascular adhesion under fluid shear force of the Lyme disease pathogen Borrelia burgdorferi. Using competitive inhibition approaches, we have determined that Borrelia vascular adhesion in vivo is dependent on glycosaminoglycans and a GAG-binding bacterial protein, BBK32, which is conserved in other bacterial pathogens. The identity of the glycosylated structures and host receptors targeted by BBK32 is unknown. As part of our characterization of the BBK32-regulated vascular adhesion mechanism, we would like to identify the glycosylated structures recognized by BBK32, in an effort to understand how this lectin mediates adhesion under shear force.

Pun , PhD Associate Professor,
University of Washington Subgroup(s): [To be Assigned]
Research interests: The focus of the research in my lab is the delivery of biologics (i.e. dna, peptides, proteins) via viral vectors, synthetic polymeric vectors, and polymer-virus hybrid vectors as well as targeted delivery of molecular imaging contrast agents for cancer detection. We have developed several sugar-based materials for delivery and cell targeting. I contributed toward the development of the first cyclodextrin-based polymers used in clinical trials for siRNA delivery [1,2]. We have also investigated the role of particle size in galactose-mediated nanoparticle targeting to hepatocytes [3,4]. In gene delivery studies, we have shown the critical role of heparan sulfate proteoglycans (HSPGs) for mediating cell uptake of polymer-coated adenovirus for transduction [5]. For cancer detection projects, glycan targets have been identified as potential biomarkers for identification of precancerous tissue in the esophagus [work in progress]. Thus, my lab has come to appreciate the importance of glycans in mediating biological processes, and we intend to further study these mechanisms in order to engineer more effective vectors for gene delivery and cancer detection. The CFG houses a valuable number of resources such as glycan array screenings and the reagent bank that can further this research.
